Objectives of the Course

Teaching microscopic examination methods.

Course Content

Introduction to microscope and microtome, smear preparation technique, bone preparation. Paraffin technique; detection solution selection, preparation and application, rules to be considered in the detection process, dehydration process, embedding and embedding errors, sectioning and points to be considered when sectioning, painting, selection of paint for the purpose, preparation of paint and paint set and different painting techniques, mounting process.

Learning Outcomes
1.Learns how to make preparations of cells, tissues and organs to be examined under the microscope.
2.Understands laboratory studies and uses basic methods.
3.Learns and applies different preparation and microscopic examination methods and understands their importance.
4.Knows the method of examination after live and detection, and understands the difference between them.
5.Knows the paraffin preparation method, applies it, and produces solutions to the problem.
6.Examines and interprets the resulting preparation at the microscope level.
7.He/she reaches a level where he/she can teach examination methods.
8.Solve problems in laboratory studies and establish relationships with different fields during interpretation processes.

Week 1 - Theoretical & Practice
Introduction to the microscope and microtome.
Week 2 - Theoretical & Practice
Introduction of experimental animals, selection and care of experimental animals in experiments.Preparation of the experimental animal for the operation, dissection, tissue and organ harvesting.
Week 3 - Theoretical & Practice
The importance of fixation, selection of fixation solutions (fixatives). Preparation of fixatives.
Week 4 - Theoretical & Practice
Points to be considered during the detection process are detection errors. The process of detecting tissues.
Week 5 - Theoretical & Practice
Dehydration and blocking process, its purpose and importance, points to be considered in these processes. Dehydration process; passing fixed tissues through a graded alcohol series. Blocking; The process of embedding tissues in paraffin.
Week 6 - Theoretical & Practice
Introduction and working principles of microtome, explanation of microtome types. Introduction and use of the microtome used in the laboratory.
Week 7 - Theoretical & Practice
The importance of sectioning, points to consider when sectioning. Taking sections of fixed and blocked tissues with a microtome.
Week 8 - Theoretical & Practice
The importance of sectioning, points to consider when sectioning (continued). Taking sections of fixed and blocked tissues with a microtome (continued) (Midterm Exam)
Week 9 - Theoretical & Practice
Staining process; stain and staining methods (I) Preparation of stains and stain set.
Week 10 - Theoretical & Practice
Staining process; stains and staining methods (II) The process of staining the resulting sections.
Week 11 - Theoretical & Practice
Stains and staining methods(III) The process of staining the resulting sections (continued).
Week 12 - Theoretical & Practice
Explanation of the mounting process and points to be taken into consideration. Mounting the painted sections with a mounting medium and turning them into permanent preparations.
Week 13 - Theoretical & Practice
Process, importance and purpose of smear preparation. Application of the smear preparation technique.
Week 14 - Theoretical & Practice
Process, importance and purpose of bone preparation. Preparation of the bone preparation.
