Objectives of the Course

The aim of this course is to equip students with practical knowledge of fundamental laboratory techniques used in molecular genetics. Students are expected to learn methods for DNA isolation, amplification, analysis, and manipulation of genetic material. Additionally, the course aims for them to acquire skills in laboratory safety, equipment usage, and experimental data evaluation.

Course Content

This laboratory course begins with an introduction to general laboratory rules and basic equipment. Fundamental molecular techniques are applied, such as DNA isolation from prokaryotic and eukaryotic cells, quantitative determination of DNA, and analysis of DNA thermal stability. DNA analysis is performed using agarose gel electrophoresis. Polymerase Chain Reaction (PCR) is used for the amplification of specific gene regions, and restriction enzyme digestion (RFLP) procedures are carried out. Applications such as plasmid DNA isolation, determination of colicin factor presence, and investigation of UV effects on bacteria are performed. Additionally, gene transfer studies are conducted using competent cell preparation and bacterial transformation techniques. The course includes practical applications designed to enable students to learn molecular genetic laboratory techniques through experience.
