Objectives of the Course

To provide information about plant cell and tissue culture techniques and to ensure the acquisition of skills for their application.

Course Content

Definition and history of tissue culture, laboratory setup, preparation of culture media, selection of media, plant growth regulators, callus cultures, micropropagation, haploid plant production, protoplast culture and somatic hybridization, somatic embryogenesis, organogenesis, production of secondary products, obtaining virus-free plants, caryopreseration of cultures, tissue culture in plant breeding.

Week 1 - Theoretical & Practice
Definition and terminology of tissue culture. Basic laboratory principles.
Week 2 - Theoretical & Practice
Tissue culture laboratory organization. Introduction to laboratory equipment.
Week 3 - Theoretical & Practice
Culture media and preparation steps, solution preparation, pH measurement.
Week 4 - Theoretical & Practice
Plant growth regulators. Stock solution preparation methods and stock solution preparation.
Week 5 - Theoretical & Practice
Organogenesis. Preparation and sterilization of basic MS culture medium.
Week 6 - Theoretical & Practice
Somatic embryogenesis. Plant material culture (PDA) for sterilization determination.
Week 7 - Theoretical & Practice
Micropropagation. Review of last week's cultures.
Week 8 - Theoretical & Practice
Protoplast culture and somatic hybridization. Review of case studies on the subject.
Week 9 - Theoretical & Practice
Embryo culture. Preparation of MS medium containing plant growth regulator.
Week 10 - Theoretical & Practice
Haploid plant production; anther culture, ovule and ovary cultures. Microscopic examination of anthers.
Week 11 - Theoretical & Practice
Cell culture and secondary metabolite production. Examination of callus cultures.
Week 12 - Theoretical & Practice
Production of virus-free plants. Examination of meristem regions.
Week 13 - Theoretical & Practice
Cryopreservation of in vitro cultures. Review of sample studies.
Week 14 - Theoretical & Practice
Tissue culture in plant breeding. Evaluation of cultures performed throughout the period.
